چکیده

Pectinases find wide application in different industrial applications, e.g., textile, food, tea, paper and pulp, vegetable oil extractions, saccharification of agricultural remains, fermented drinks. Three fungal strains were screened for pectinase production. Results indicated that all are capable producing a enzyme. Penicillium chrysogenum MF318506 showed the highest activity (0.214 U/ml) on recommended medium at 30 ◦C 150 rpm, 6 days. Different wastes (orange peel, banana potato pomegranate wheat bran, rice bran) as carbon source. The maximum 0.48U/ml was obtained from Orange peel selected further studies. Screening important components production by Plackett-Burman design (PBD) 1.057 U/ml achieved with 3 % orange 2.0 g/l (NH4)2SO4, 6.0 peptone, KH2PO4, K2HPO4, 0.1 MgSO4.7H2O, 0.5 KCl FeSO4. Finally, Box-Behken (BBD) which uses response surface methodology (RSM) applied to optimization factors using PBD. 1.292U/ml observed 2.5% 4.5 peptone. Statistical optimization, enhanced 6.04 folds waste substrate. produced has potential applications fields.

Statistical Optimization of a-Galactosidase Production in Submerged Fermentation by Streptomyces griseoloalbus Using Response Surface Methodology

a-Galactosidase production by a novel actinomycete strain Streptomyces griseoloalbus in shake flask culture was optimized using response surface methodology. Screening of variables to find their relative effect on a-galactosidase production was done using Plackett-Burman design. Out of the eleven factors screened, salinity, magnesium sulphate and temperature were found to influence the enzyme p...

Invertase production by Penicillium chrysogenum and other fungi in submerged fermentation.

A study was made of Penicillium chrysogenum and some other fungi to determine the relative distribution of intra- and extracellular invertase produced by them in submerged fermentation. The proportion of each type of enzyme varied with the organism and the period of fermentation. More of the enzyme initially bound to the mycelium was released into the medium with the progress of fermentation. D...

Application of Response Surface Methodology (rsm) – Ccd for the Production of Laccases Using Submerged Fermentation

Laccases are multi-copper oxidases widely distributed in plants, animals, insects and microorganism. These enzymes catalyze the one-electron oxidation of a wide range of organic and inorganic substances, including mono, di and poly phenols, aminophenols, methoxy phenols, aromatic amines and ascobate with the concomitant four electron reduction of oxygen to water. Laccase has the focus of much a...
